Upright / Compound Microscopes

Upright biological compound microscopes are used for observing thin, transparent, slide mounted specimens under magnifications typically ranging from 40X to 1000X.  They are equipped with a bottom light which provides transmitted illumination through the specimen.  Typical specimens suitable for observation using this type of microscope are histology sections, thin sections of plant material, blood, cytological specimens, crystals, emulsions and transparent parts of organisms such as insects.

Compound microscopes have the option of one eyepiece to view through (monocular microscopes), two eyepieces for more comfortable viewing (binocular microscopes) or two eyepieces plus a third port for attaching a camera (trinocular microscopes).

There are a number of different illumination techniques available with compound microscopes, including brightfield illumination, phase contrast, DIC, darkfield and fluorescence. Phase contrast provides additional contrast for viewing unstained specimens that are not possible to see via brightfield illumination, for example living cells.

DIC microscopeCompound Microscopes with DIC (Differential Interference Contrast) Illumination

DIC illumination is the next step up from phase contrast that allows thicker samples to be observed (a limitation of phase contrast). It can be used in conjunction with fluorescence illumination, where DIC is used to locate a specific region on the specimen before switching to fluorescence for further examination.

For viewing specimens that have been tagged (dyed) with a fluorescent label. Most often used for research.

Darkfield microscopeCompound Microscopes with Darkfield Illumination

Darkfield is an illumination technique used to view samples that do not image well under normal illumination. Light from the condenser is blocked by a stop and forced around the edges to illuminate the sample via oblique angles, creating a bright image of the specimen on a dark background. This creates a particularly eye-catching image and is popular with microscopy photographers.
